Born and raised in Vandalia, Illinois, June Squibb trained at the Cleveland Play House and the highly regarded HB (Herbert Berghof) Studio in New York City. She began her career in musical theater and got her first big break when she landed the role of Dulcie in the 1959 Off-Broadway production of The Boyfriend. Her Broadway debut came the following year, in the original production of Gypsy starring Ethel Merman, playing the role of a stripper named Electra. June made her feature film debut in 1990 in Woody Allen's Alice, starring Mia Farrow. Roles in Scent of a Woman (1992), The Age of Innocence (1993) and Meet Joe Black (1998) followed.

In 1995 June played her first television role with a guest appearance on the TV series Law & Order. In 1999 she played another guest role on the series. In 2003 she landed a recurring role as Louise Flower on the drama series Judging Amy and from 2005 to 2007 she played a recurring role as Jennifer Love Hewitt's "Grandma" on the supernatural series The Ghost Whisperer. More recently, she joined the cast of Girls, playing Hannah Horvath's grandmother.

June often returns to the theater and in 1995 played a role in Sacrilege on Broadway alongside Ellen Burstyn.

In 2014, June received her first Oscar nomination for her role as Bruce Dern's wife Kate in the major motion picture Nebraska.

Her recent projects include the Blythe Danner drama I'll See You in My Dreams (2015), the Steve Martin, Diane Keaton and John Goodman comedy Love the Coopers (2015), the Jesse Plemons and Molly Shannon drama Other People (2016), seven episodes of the Golden Globe-nominated series Shameless in 2016, and the comedy Table 19 (2017) with Anna Kendrick.
